奶牛乳头状瘤病理学诊断及病原分子检测研究

摘要

2012年6月新疆沙湾县某奶牛养殖场爆发疑似牛乳头状肿瘤病,为了确诊该病,本研究采取皮肤病变组织进 行病理切片检查;设计牛乳头状瘤病毒简并引物 BPVP1/BPVP2对皮肤肿瘤组织样品进行 PCR 扩 增,扩增 产 物 克隆至pMD18-T载体中进行测序、序列分析及遗传进化树构建。结果表明:遗传进化树分析显示新疆沙湾县爆发 的牛乳头状肿瘤病是由2型牛乳头瘤病毒所引起,与 BPV-1、BPV-13进化关系最近。本研究为新疆地区牛乳头状 瘤的科学防控提供了理论依据。

Abstract

InJune2012,asuspectedcutaneouspapillomatosisbrokeoutinadairyfarminShawan,Xinjiang.Inordertodiagnose thedisease,skinlesionswerecollectedfortissuehistopathologicalexamination.ApairofprimerBPVP1/BPVP2wasdesigned todetectPBVinskintumortissuebyPCRtechnique.TheamplifiedfragmentwasclonedintopMD18-Tvector,andthense- quencedforphylogenetictreeanalysis.Theresultshowedthattheoutbreakoftumordiseaseiscausedbybovinepapillomavirus type2,whichwasmostcloselyrelatedtoBPV-1andBPV-13.Thisstudywasprovidedatheoreticalbasisforthescientificpre- ventionofbovinepapilloma.
