摘要

以金电极为电极材料,纳米铂/普鲁士蓝复合物膜为固载基底,癌胚抗体(anti-CEA)为模板免疫蛋白质分子, 研制成一种高灵敏电流型癌胚抗原免疫传感器。该传感器以辣根过氧化氢酶(HRP)代替牛血清白蛋白封闭非特 异性结合位点,放大了电流信号,提高了电极的灵敏度,并采用循环伏安技术对电极的制备过程进行了研究。在优 化的实验条件下,该传感器响应的峰电流值与癌胚抗原浓度在1.0~80.0ng/mL的范围内保持良好的线性关系, 检测限为0.4ng/mL

Abstract

Anewamperometricimmunosensorforthedeterminationofcarcinoembryonicantigen(CEA)wasdevelopedbymeans ofimmobilizingcarcinoembryonicantibodyontothenano-Pt/Prussian-blue-modifiedcompositefilm.Theresearchsubstituted Horseradishperoxidase(HRP)forbovineserumalbumin(BSA)toblocknonspecificbindingsitesandamplifythecurrentsig- naloftheantigen-antibodyreaction.Cyclicvoltammetry (CV)wasemployedtocharacterizetheassemblyprocess.Underthe optimizedconditions,theresponsecurrentoftheproposedimmunosensorhasagoodlinearrelationshipwithCEAintheconcen- trationrangesfrom1.0to80.0ng/mLwithadetectionlimitof0.2ng/mL
