The Experimental Study of Interleukin-18 Effects in Rat Retinal Neovascularization

为探讨白介素-18对大鼠视网膜中血管内皮生长因子(VEGF)及新生血管形成的影响。采用50只7 d龄清洁级SD大鼠幼鼠随机分为5组,即空白对照组、高氧未处理组、生理盐水实验组、高浓度实验组和低浓度实验组,建立高氧诱导的新生大鼠产生视网膜新生血管的动物模型。分别对3个给药组双眼玻璃体腔内注射生理盐水、IL-18(50 ng/μL、50 ng/μL)1μL。免疫组织化学染色法观察VEGF在视网膜各层的表达,应用CD34计算突破视网膜内界膜的新生血管内皮细胞核数,比较各组间差异。结果显示,(1)HE染色显示,各实验组均有较多新生血管内皮细胞核突破视网膜内界膜进入玻璃体腔内,但高浓度实验组及低浓度实验组明显低于生理盐水实验组及高氧未处理组,并且高浓度实验组低于低浓度实验组。(2)VEGF在各组视网膜中均有表达,在高氧组表达明显增加,IL-18给药组表达相对较弱,空白对照组最弱。(3)空白对照组极少见增生的毛细血管,其余各组发现较多CD34标记的增生的毛细血管,高浓度实验组、低浓度实验组相对较少,且组间比较差异有统计学意义(P<0.05)。由此可知,IL-18玻璃体腔内注射可抑制血管内皮细胞的增殖及迁移,从而抑制视网膜新生血管的形成。
To investigate the effects of IL-18 on retinal neovascularization and the expression of VEGF on mice.Fifty seven-day-old neonatal SD mice were randomly divided into five groups:control group、NaCl group、hyperxia experimental group,low concentration experimental group and high concentration experimental group.Retinalneovascularization modelswere induced by hyperxia in newborn m ic.The mice in injected group were respectively treated with intravitreal injection of Nacl、interleukin-18(10 ng/μL and 50 ng/μL)1 μL.The expression of VEGF at layers of retina was detected by immunohistochem istry.The numbers of the endotheliocyte nuclei of neovascularization which broke through the retinal inner lim itingmembranewere calcu-lated by CD34 in order to evaluate inhbiting effects of the Bevacizumab on the retinal neovascularization.(1)The number of endotheliocyte nuclei of neovascularization was small in control group,but in hyperxia experimental group,low concentration experimental group and high concentration experimental group,there are much more endotheliocyte nuclei and the low concentration experimental group was more than in high concentration experimental group.(2)The expression of VEGF was found at all layers of retina in each group,significantly increased in hyperxia experimental group,relatively decreased in high and low dose experimental group,and was the lowest in control group.(3)The expression of neovascularization by CD34 was found at all layers of retina in each group,significantly increased in hyperxia experimental group and NaCl group,relatively decreased in high and low concentration experimental group,and was to the lowest in control group.There was significant differences for low and high concentration experimental group compared(P<0.05).Interleukin-18 can inhibit the endotheliocyte nuclei proliferation and migration,effectively inhibit the retinal neovascularization.
