摘要

目的 探讨小鼠肝细粒棘球蚴感染模型中 TLR2 表达及其对肝组织的影响。方法 选取 40 只 C57BL/6 小鼠随 机分为对照组、感染组,感染组以 0. 1mL 原头蚴悬液( 含原头蚴 5 000 个) 注射于肝脏被膜下建立继发感染肝细粒 棘球蚴模型,对照组注射等量 PBS。感染后 3、7、14、21 d 处死小鼠,摘取肝脏,观察肝脏大体形态,行 HE 染色, qRT-PCR、免疫组化检测肝脏中 TLR2 的表达。结果 与对照组相比,模型组小鼠一般情况良好,感染率 100%,随着 感染时间延长,肝组织表面囊型物增大并凸出与肝脏表面,HE 染色可见肝脏病灶出现炎性细胞浸润和纤维组织增 生等多种病理改变。模型组的 TLR2 基因及蛋白相对表达量显著增高,且随着感染时间的延长,阳性表达呈持续高 表达。结论 细粒棘球蚴感染宿主后,肝组织 TLR2 表达增高,出现明显炎症改变。

Abstract

Objective To investigate the expression of TLR2 in mouse liver echinococcus granulosus infection model and its effect on liver tissue.Methods Forty C57BL/6 mice were randomly divided into a control group and an infected group.The infected group was injected with 0. 1 mL of protoscolices suspension ( including 5000 protoscolices) under the liver capsule to establish secondary infection of Echinococcus granulosus.The model and the control group were injected with the same amount of PBS.The mice were sacrificed on days 3,7,14,and 21 after infection,and the livers were harvested to observe the general morphology of the livers,followed by HE staining, qRT-PCR,and immunohistochemistry to detect TLR2 expression in the liver.Results Compared with the control group,the mice in the model group were generally in good condition with an infection rate of 100%.With the prolonged infection time,the cysts on the surface of the liver tissue increased and protruded from the surface of the liver.HE staining showed inflammatory cell infiltration in liver lesion and fibrous tissue hyperplasia and other pathological changes.The relative expression of TLR2 mRNA and protein in the model group increased significantly,and as the infection time prolonged,the positive expression continued to be high.Conclusion After Echinococcus granulosus infection of the host,the expression of TLR2 in liver tissue increased,and obvious inflammation changes appeared.
